Names of new reads

Post by jstamm » Wed Feb 13, 2008 8:40 pm

I just downloaded some of the new reactions that we called. The reads have names like these:

I know that the _t suffix indicates 4:1 chemistry, and that _t1 indicates that the read was performed with primer 1. What do a and b indicate? Are these multiple identical runs? All the chromats look pretty bad, so I'm guessing that's what it is, but I would like confirmation.

a and b names for oligos

Post by cshaffer » Thu Feb 14, 2008 12:15 am

Since each project is claimed twice some of the clones are being worked on simultaneously by two people. If both people order oligo's the same week we can have two different oligo's with the same number. To split these out we add an "a" and "b" label. We are continuing to work on the script that handles this, in future weeks we will probably add something to indicate faculty name to avoid confusion.

One set should match the oligo's you called. Since this is a new script, the possibility for mistakes is high, so if something looks like it is labeled incorrectly it probably is. So take these "a" and "b" labels for what they are which is our first attempt to explicitly deal with the issue of oligos coming from two projects.
